(WKN A0RM18, AMT, M20 Frankfurt Exchange) announced today that proprietary Blood Cell Morphometric Method (BCMM) technology was implemented using inspection system Prototype AMT 007 to find and measure lymphocytes nuclei and cytoplasm dimensions. This data collected by motorized microscope system is key lymphocytes statistics used for determination of cancer disease and its stage. Lymphocytes are quite common in the blood: 20-40%, 8-10 µm in diameter and generally they are smaller than the other leukocytes but they are still a few larger than red cells. The cytoplasm is transparent. The nucleus is round and large in comparison to the cell and it occupies most of it. Our Prototype is fully automated system consists of Nikon Eclipse 50i microscope with Ludl BioPoint 2 motorized XY stage and motorized focus Z stage with linear encoder to achieve focus resolution up to 0.1 micron. Extremely high positional accuracy for the focusing lens is essential to achieve accurate measurement results fully correlated with original manual inspection results. It is also important for production system collecting images with continuous movement of the XY stage. Software choice for quick development of prototype system was Media Cybernetics, Inc. IMAGE-PRO bio- medical software. It is well known by FDA and bio-medical research scientists. Motorized XYstage moves a glass smear under the microscope and digital camera (QICAM) captures one by one frame located close to each other. Distance between neighbor frames is approximately 30% of frame width and height to avoid repeatable measurement of the same cells located on the borders. Nuclei color (usually dark blue) is unique for the image. The first part of inspection is to find nuclei color objects (or BW level objects) satisfying the dimensional limits for nuclei. Only lymphocytes with oval single nuclei 4 to 12 micron in size will be considered in collected data. Roundness characteristics of the nuclei help us to reject other types of cells (different than lymphocytes). Statistics shows that 15 to 20% of lymphocytes will be detected on the borders of the frame and system moves these cells to the center of the camera Field of View (FOV). More complicated task is to find a cytoplasm, but we are looking for one in the area concentric to the nuclei center. This eliminates possible foreign cells located in proximity to the nuclei. Positional information of cells collected and stored automatically on the PC hard drive. Inspection of the smear can be repeated with the same or higher resolution camera and even higher magnification lens based on positional information. Our Prototype system can also visually sort cells images and eliminate all unnecessary data and algorithms can be used in next generation of our production system.
